A microtome knife guard.
نویسنده
چکیده
The base sledge microtome presents a hazard to the operator due to the length of blade that is exposed. Manipulation of the carriage requires hand movements close to the knife edge and serious lacerations may result. A simple, effective guard has been devised, suitable for knives which have screw holes for handles at both ends. It consists of two perspex end-pieces (Fig. I), attached to the knife ends by screws, and linked by a metal bar (Fig. lb). The bar guards the whole length of the knife, with minimal interference to adjusting its position in the clamps. In addition the knife edge is protected from accidental damage should the tissue chuck be raised too high. When suitably aligned, the guard acts as a 'level finder' for the top of the tissue block.

The regular production of a satisfactory edge on a microtome knife for the cutting of sections as thin as 2 u, which are free from scores and other blemishes, is a serious technical problem in histological laboratories. متن کاملSome Effects of the Microtome Knife and Electron Beam on Methacrylate-embedded Thin Sections
A technique for the examination of specimens at low electron beam intensity has been presented. Sections micrographed with this technique showed numerous knife scratches and frequently contained bands running parallel to the knife edge. Banding with an average spacing of 0.2 micro appeared to result from periodic distortion produced by impact of the knife. At the beam intensities customarily em...
متن کاملFrozen Thin Sections of Fresh Tissue for Electron Microscopy, with a Description of Pancreas and Liver
A simple method has been developed that allows frozen thin sections of fresh-frozen tissue to be cut on a virtually unmodified ultramicrotome kept at room temperature. A bowl-shaped Dewar flask with a knifeholder in its depths replaces the stage of the microtome; a bar extends down into the bowl from the microtome's cutting arm and bears the frozen tissue near its lower end. When the microtome ...
متن کاملStaining Differences in Cell Nuclei
If the parenchymal cells of the liver of the rat, or certain other cells, are stained with a modified Mallory stain or with iron haematoxylin, the nuclei are stained in two different ways. This effect may be observed after various fixatives. Direct microscopical observation shows that when part of a nucleus is cut away by the microtome-knife, the part of it remaining in the section stains diffe...
متن کاملA Diamond Knife Holder for Microtomes Designed to Take Glass Knives
Diamond knives for cutting thin sections for electron microscopy were first introduced by Ferngndez-Mor~n (1) and have been perfected subsequently by him (2). Such knives, as distributed, are embedded by means of a special alloy in a rectangular metal mounting which usually measures 11 x 4.5 x 1.5 mm. (2). A holder for the metal mounting, complete with collecting trough, has been described (2),...
